Along with other structurally related neurotrophic factors NGF, NT-3 and NT-4, BDNF binds with high affinity to the TrkB kinase receptor. It also binds with the LNGFR (for low-affinity nerve growth factor receptor, also known as p75). BDNF promotes the survival, growth and differentiation of neurons.It serves as a major regulator of synaptic transmission and plasticity at adult synapses in many regions of the cns.BDNF expression is altered in neurodegenerative disorders such as Parkinsons and Alzheimers disease.DescriptionRecombinant Human Brain-derived neurotrophic factor produced in E. coli is a non-glycosylated polypeptide chain containing 119 amino acids.Bioactive BDNF is a non-covalently linked homodimer and having a molecular mass of 27kDa.Quality ControlBiological activity: The ED50 of rHuBDNF as determined by cell proliferation assay using the TrkB transfected BaF3 mouse pro-B cell line was 1-6ng/mL. Purity: Greater than 98.0% as determined by: (a) Analysis by RP-HPLC. (b) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel. Amino-Acid Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-His-Ser-Asp-Pro . Endotoxin: Less than 0.1ng/μg (1EU/μg) of hBDNF as determined by LAL test. FormulationSterile and Lyophilized aliquots with a 0.2um filtered protein solution containing 20mM PB pH8.0, 400mM NaCl.ReconstitutionIt is recommended to reconstitute the lyophilized rHuBDNF in sterile 18MΩ-cm H2O not less than 100μg/ml, which can then be further diluted to working solutions.Storage Lyophilized rHuBDNF although stable at room temperature for 3 weeks, should be stored desiccated below -18°C, which is stable for up to twelve months from date of receipt. Upon reconstitution rHuBDNF should be stored at 4°C between 2-7 days and aliquots of reconstituted samples are stable up to 3 months below -18°C for future use.Please avoid freeze-thaw cycles.
