抗原受体基因的重排发生于T/B淋巴细胞个体生成时期，这种重排产物对于每一个淋巴细胞来说具有序列上和长度上的唯一性。因此，可以应用PCR的方式通过检测抗原受体基因位点上独特的V-J区域基因重排来鉴别不同来源的淋巴细胞数量。TheIdentiClone™ Bcell Gene Clonality Assay使用多条引物靶定immunoglobulin lambda、delta、kappa light chain gene的保守区域。这种方式能够检测绝大多数克隆性的B细胞恶性肿瘤。同时检测结果可以用多种方法进行分析，如电泳，荧光等。
InVivoScribe的IdentiClone™检测是对基于PCR的克隆性检测新方法。这些标准化的检测都经过精心优化，使用多重混和试剂进行阳性对照和阴性对照，并经过大量验证，检测超过400个经欧洲/美国淋巴瘤分类Revised European/American Lymphoma (REAL) 的临床样品。
基因重排同样可以用Southern Blot分析方法来检测。尽管SB方法可行度很高，但还是越来越多的被PCR检测方式所取代，因为PCR方法更高效和灵敏。而且PCR方法相对简单，需要的劳动力和样品量都更少。另外PCR方法可以检测从石蜡包埋样品中分离得到的DNA，而SB方法却不行，因为在分离过程中DNA常常是会降解的。
