The chemical you are studying associates with cis element in CMV promoter, it may affect promoter transcription activity, but has nothing to do with mutations of transgene. This is why the company told you that the compoany was not able to make it.

If you want to know whether the chemical you are studying can modify the transgene product function, you need to know first if the chemical stimulates or suppresses that function or not, then you should elucidate how the chemical affects transgene product function by examining function activity of mutant forms of transgene product.

首先非常感谢您帮助！我研究的是UGT1A1基因，苯巴比妥对它有诱导作用已经被证明了，那我现在想研究的是苯巴比妥对UGT1A1基因不同突变型的诱导作用是否有差异，（这个突变在编码区）我想通过构建UGT1A1不同突变型的慢病毒载体来研究，但是苯巴比妥的作用位点在UGT1A1基因启动子上游的苯巴比妥反应增强元件，所以我能把这个苯巴比妥反应增强元件和UGT1A1基因编码区一起构建吗？或者有没有别的办法呢？还请指教！
